So, they are bigger because they are each made individually to a pattern designed by the Emperor (Maybe with his direct DNA infusion, but unconfirmed as far as I know.). To learn more, any Custodes Codex, Watchers of the Throne, and Valdor are all good reading, and old Codexes are cheap.

Gene Seed is a very specific thing. It's another name for the Proginoid Glands in Space Marines. Every Marine has 2 sets. One is removed at maturity, the other at death. Custodes simply do not have or need these organs, thus, not created with Gene Seed. GK do have these organs.

Custodes are not made with Gene Seed, fyi.
